Genetic dissection of Drosophila adenylate cyclase.

چکیده

The Drosophila learning mutant rutabaga is missing calcium/calmodulin activation of adenylate cyclase (EC4.6.1.1). The mutation was mapped at a finer resolution to X chromosome bands 12F5-7. By comparing wild-type and mutant cyclase activities, the relative responsiveness of the calcium-sensitive and calcium-insensitive components to different ligands could be determined; the calcium-sensitive fraction of the total cyclase activity was significantly less responsive to guanyl nucleotides and monoamines. The results suggest that the component of cyclase activity that is stimulated by calcium/calmodulin, possibly a genetically distinct catalytic subunit, is not coupled to the G subunit or the G subunit/monoamine receptor complex.

E VALUATION OF ADENYLATE CYCLASE ACTIVITY IN MITRAL VALVE PROLAPSE

The term mitral valve prolapse (MVP) is used for a particular subset of patients with hyperadrenergic dysautonomia. It occurs when part of a leaflet or both leaflets of the mitral valve extend above the plane of the atrioventricular junction during ventricular systole. The adenylate cyclase activity in MVP dys-autonomia was studied by extraction of enzyme from the erythrocytes from 62 norma...

Genetic Dissection of Age-related Memory Impairment in Drosophila

Age-related memory impairment (AMI) is an important phenotype of brain aging. Understanding the molecular mechanisms underlying AMI is important not only from a scientific viewpoint but also for the development of therapeutics that may eventually lead to developing drugs to combat memory loss. AMI has been generally considered to be an overall or nonspecifi c decay of memory processes that resu...

Rescue of the learning defect in dunce, a Drosophila learning mutant, by an allele of rutabaga, a second learning mutant.

rutabaga1 (rut1), a Drosophila learning mutant, has adenylate cyclase (EC 4.6.1.1) with reduced basal activity and the absence of calcium/calmodulin-stimulated activity. A second learning mutant, dunce, is defective in cyclic AMP degradation due to decreased or absent phosphodiesterase activity. These opposing biochemical defects allow rut1 to partially suppress the female sterility caused by e...

A novel synaptic transmission mediated by a PACAP-like neuropeptide in drosophila

Neuropeptide-mediated transmission was analyzed at Drosophila larval body-wall neuromuscular junctions. Focal application of vertebrate pituitary adenylyl cyclase-activating polypeptide (PACAP38) to the neuromuscular junction region triggered two temporally distinct muscle responses: an immediate depolarization followed by a large enhancement of K+ current. This late enhancement occurred many m...

Viability, longevity, and egg production of Drosophila melanogaster are regulated by the miR-282 microRNA.

The first microRNAs were discovered some 20 years ago, but only a small fraction of the microRNA-encoding genes have been described in detail yet. Here we report the molecular analysis of a computationally predicted Drosophila melanogaster microRNA gene, mir-282. We show that the mir-282 gene is the source of a 4.9-kb-long primary transcript with a 5' cap and a 3'-poly(A) sequence and a mature ...
